Mutations in fibroblast growth factor receptors (FGFR) can affect receptor tyrosine kinase (RTK)-Ras signaling with profound biological consequences in cell growth, development, and differentiation. A large number of mutations in FGFRs have been discovered in cancer, and a few have been described to expand in the male germline with paternal age. In spite of the severe consequences that mutations have in these receptors, little is known about the biochemical and biophysical changes in the signaling caused by the mutations. In this project, we propose to discover mutations in the Fgfr3 gene expanding in the male germline and characterize their biochemical and biophysical consequences.
